MK Dons (16/1) have been drawn at home to League One rivals Charlton in the Johnstone’s Paint Trophy, in what looks the tie of the second round.
Swindon, who defeat holders Southampton in round one, have been rewarded with a home tie against League Two Torquay in the Southern section west.
Last year’s beaten finalists Carlisle will also be at home in round two, with Port Vale the visitors to Brunton Park.
There’s a London derby to look forward to at Brisbane Road, as Leyton Orient face League One’s bottom club Brentford.
Favourites Sheffield Wednesday (Brentford 12/5, Draw 12/5, Sheff Wed 11/10 - Sunday Match Betting) will fancy their chances of progression having been drawn to face League Two Chesterfield.
The second round ties will take place in the week beginning October 4.
Northern section (west): Accrington v Stockport, Macclesfield v Crewe, Bury v Shrewsbury, Carlisle v Port Vale.
Northern section (east): Burton v Rotherham, Hartlepool v Bradford, Sheffield Wednesday v Chesterfield, Huddersfield v Peterborough.
Southern section (west): Bristol Rovers v Aldershot, Swindon v Torquay, Cheltenham v Plymouth, Hereford v Exeter.
